Surgical procedures

Vascular indwelling catheters
Indwelling venous catheters can be used for various infusions including tracers, metabolic substrates, peptides, drugs, etc. Arterial catheters are used for blood sampling in a non-stressing condition. Catheters are placed under anesthesia, after loss of pedal and corneal reflexes is assured. Mice are allowed to recover for 4-7 days and only mice that have regained >95% of their preoperative weight are studied.

Number of mice will depend on the experimental protocol and should be discussed. Subsequent experiments should be best performed in our facility.

Intracerebroventricular (ICV) injections
Cannulation of cerebral ventricle allows for central infusions of metabolic substrates, receptor ligands, drugs or recombinant viruses (lentiviruses, adenoviruses). A stainless steel cannula (Plastics One, Roanoke, Virginia) is implanted in the right lateral ventricle of anesthetized mice secured in a stereotactic frame. The animals are then allowed to recover for at least 4 days and only mice that have regained >95% of their preoperative weight are studied. Placement of cannulas is optimized for each strain by histological analysis of frozen brain sections, and verified functionally in each tested mouse by stimulation of drinking behaviour with ICV injection of angiotensin.
